Class of drug

  • Protein-tyrosine kinase inhibitor.

Uses

Action

  • Selectively inhibits the mutated form, in the juxtamembrane (JM) region, of the c-kit receptor.
  • Also inhibits the Platelet Derived Growth Factor (PDGF) and the Fibroblast Growth Factor Receptor (FGFR3).

Indications

  • Treatment of non-resectable canine mast cell tumors Skin: mastocytoma (Grade 2 or 3) with confirmed mutation of the c-kit tyrosine kinase receptor.
  • For any mast cell tumor treatable by surgery, surgery should be the first choice of treatment.

Other reported reactions

  • Mild to moderate gastrointestinal reactions (diarrhea and vomiting) with a mean duration of approximately 21 and 9 days respectively.
  • Mild to moderate alopecia with a mean duration of approximately 26 days.
  • Severe renal toxicity may occur in dogs with renal disorders at start of treatment (including high blood creatinine level Blood biochemistry: creatinine or proteinuria Proteinuria ).
  • Mild to severe anemia (aplastic/hemolytic) with a mean duration of approximately 7 days.
  • Protein-loss syndrome (mainly due urinary protein losses).
  • Mild or moderate neutropenia with a mean duration of approximately 24 days.
  • Increase in aminotransferases (ALT or AST) with a mean duration of approximately 29 days.
  • Other commonly observed adverse reactions were in most cases mild or moderate:
    • Lethargy and asthenia (mean duration approximately 8 and 40 days respectively).
    • Decreae in appetite or anorexia (mean duration 45 days and 18 days respectively).
    • Cough (mean duration 23 days).
    • Lymphadenopathy Lymphadenopathy (mean duration 47 days).
    • Edema (mean duration 7 days).
    • Lipoma (mean duration 53 days).
  • In case of adverse events following overdose, treatment should be discontinued until resolution, and then resumed at the recommended therapeutic dose level.
